We have discussed use of the host file for ad blocking before.
Sophisticated ad blockers like AdMuncher and AdBlock eliminate the need to use the host file to accomplish the same thing.
While you CAN use the host file for blocking, loading huge lists into it can degrade performance as it does consume memory and processing.
Some ad blockers ADD to the host file and store their list there.

A good use of the host file is blocking unwanted ads in applications other than your internet browser, like PalTalk, where there is no ad blocker integration like you have in your web browser.
We previously detailed how to block annoying popups, banners, and other gunk you get in PT by entering these IPs into your host file.
